Pennsylvania Station, in Midtown Manhattan, is the busiest of the LIRR's three western terminals, serving almost 500 daily trains. Like the station itself, it is reached via the Amtrak-owned East River Tunnels (the only LIRR-used trackage not owned by the LIRR) from the Main Line in Long Island City.

The LIRR service uses the 3 & 4 lines, which run under 33rd Street from the tunnels to Penn Station, and which usually use tracks 20 & 21 in the station served by platform 11 on the extreme north-side of the station. Access is gained from the Connecting Concourse, which in turn has its own separate stairway and elevator access from an entrance building on the south side of 34th Street just west of Seventh Avenue.

Connections are available to the New York City Subway's 34th Street – Penn Station (IRT Broadway – Seventh Avenue Line) (1 2 3 trains) and 34th Street – Penn Station (IND Eighth Avenue Line) (A C E trains) stations which are next to the terminal. It also connects LIRR with Amtrak and New Jersey Transit trains.

Food and shopping can be had at Penn, and the LIRR concourse is one of the better locations for this.
