The New 42nd Street Studios building is a state-of-the-art, 10-story rehearsal studio/office/theater “complex” comprised of the New 42nd Street Studios and The Duke on 42nd Street theater, housed within the American Airlines Theatre complex. Opened in 2000, the New 42nd Street Studios is considered the premier rehearsal space in all of New York City, serving a vast and diverse group of nonprofit dance, theater and opera groups, as well as commercial Broadway musicals and plays. Designed by Platt Byard Dovell Architects, the New 42nd Street Studios building itself was awarded the New York State Association of Architects/AIA Excellence in Design Award.

The original plan when renovating the former Selywin Theatre was to incorporate the Italian-Renaissance stye facade of that theater's office portion which faced 42nd Street, but the historic facade collapsed on December 30, 1997. Instead, a new, modern facade was designed by Charles Platt and Ray Dovell (see main American Airlines Theatre entry for description).

Five of the floors contain 14 studios, with the other floors housing the 199-seat Duke Theatre and the administrative offices of The New 42nd Street and several other non-profit organizations.
