Located within the Alexander Hamilton U.S. Customs House is the Commisioner's office on the first floor. It was designed c. 1905 by Tiffany Studios. Fully panelled in oak from floor to ceiling, the office is divided by an elaborately-carved Renaissance-inspired screen which is pierced by a doorway marked by a segmentally-arched hood. Above the oak walls were placed ten paintings of 17th-century ports selected because of their relation to the settlement and commerce of the Dutch and English colonies. The paintings and probably all the painted decorations in the vaulting of the Lobby were by the muralist Elmer E. Garnsey.

It is typically only open to the public for the Open House New York festival each year in October. The interior was used as a filming location for Spike Lee's "Inside Man" (2006). More recently, it was used for the Saturday Night Live skit "Red Flag Perfume by Chanel" in 2013 and for the Michael Almereyda film "Tesla" (2020).
