Holmes Beach is a coastal city on the Gulf of Mexico in Manatee County, in the U.S. state of Florida. Holmes Beach sits on two barrier islands known as Anna Maria Island and Key Royale. Anna Maria Island is shared with the neighboring cities of Anna Maria and Bradenton Beach.

The area was first homesteaded in 1896, and by 1902 had established a post office. The city was not incorporated until 1950, when it was named Holmes Beach for Jack Holmes, a developer who arrived on the island after World War II and developed 600-acres of the community.

Holmes Beach has a total area of 1.7 sq mi (4.5 km2), of which all but 0.1 sq mi (0.3 km2) is on land. In the 2000 census, the city had a population of 4,966.
