The hospital was founded by Dr. Jose V. delos Santos, Sr. in 1949, acknowledged as the father of Philippine Orthopedics. It started as a small clinic with 30 beds and was complemented by a staff of 20. They first specialized in the treatment and rehabilitation of patients with bone disorders.

After making a mark in the medical field, the clinic expanded in 1973 and changed its name to De Los Santos General Hospital that could accommodate 150 bed-ridden patients. It then started entering the fields of Pediatrics, Obstetrics & Gynecology, Medicine and Surgery.

The hospital eventually went into education by opening a school of Nursing & Midwifery in 1975 and became a medical training institution in 1985.

Today, it has decided to focus on its core expertise by concentrating on providing health care. To do this, it has entered into a partnership with STI, a well-known computer school in the country to form the De Los Santos - STI College Inc.
