The county was formed in 1752. The original county consisted of present day Orange, Durham, Caswell, Alamance, Chatham, Person and parts of Guilford, Randolph, Rockingham, Lee, Granville and Wake counties. Over the years, parts of the county were carved off to form the modern counties. Its county seat is Hillsborough.

The southern half of the county is dominated by the growth of the college towns of Chapel Hill and Carrboro, which will eventually fill the county south of I-40. The growth is buffered to the north by the vast forest tracts owned by Duke University and by the Jordan Lake watershed to the southeast. The north half of the county is largely rural with an established farming community. In the middle is the slowly growing county seat of Hillsborough.
