The museum--at 36-11 35th Avenue, between 36th and 37th Streets in Astoria--may be off the beaten track so far as the standard New York City cultural circuit is concerned, but its site is a critical one in film history. The building is part of a national historic landmark, the mammoth complex now known as the Kaufman Astoria Studios. It was built in the early 1920's as the sound stages for Famous Players Lasky Corporation, which eventually became Paramount Pictures. The main building of the complex, next door to the museum, is the sixth largest sound stage anywhere, and the largest on the East Coast; it was once one of the most active sound stages in the world.

In 2011, the museum was expanded with a new wing which was designed by Leeser Architecture with lighting by L’Observatoire International. It now includes a 267-seat theater and a 68-seat screening room along with the Ann and Andrew Tisch Education Center.
