The building was constructed in 1930 as a Safeway grocery store, with a Pinder's Drug Store attached. It was designed by J. Melrose Morrison, originally from Scotland and whose early architectural work is rare. It was one of six stores built in Saskatoon at the same time, with a distinct Spanish-tile trim. The Mayfair Drugs location is one of only two surviving buildings of its kind - the other is Pleasant Hill Bakery.

Pinder's was renamed Mayfair Drugstore in 1939. It continued to operate out of this location until 1953, when it moved one block east and to the south side of 33rd Street - ironically, to the lot where Safeway had originally planned to build its store. Safeway moved out in 1967, and the building remained vacant until 1970.

The need for Mayfair Drugs to expand motivated its owners to purchase the former Safeway building. The dividing wall between the original drug store and grocery store was demolished, and Mayfair Drugs moved back to its original location. It has been there ever since.
